Methylation modified peptides, also known as methylation labeled peptides, are a type of post-translational modifications (PTMs) of proteins that participate in almost all cellular life processes and play an important regulatory role. Proteins covalently bind methyl groups to specific amino acid residues under the catalysis of methyltransferases. Methylation is a reversible modification process catalyzed by demethylases.

Research has found that the common amino acids involved in methylation/demethylation are lysine (Lys) and arginine (Arg). Studies have shown that histone lysine methylation modifications perform various biological functions, such as maintaining and differentiating stem cells, X chromosome inactivation, transcriptional regulation, and DNA damage response, mainly affecting chromatin condensation and inhibiting gene e­xpression. Histone arginine methylation plays an important role in gene transcription regulation and can affect various physiological processes of cells, including DNA repair, signal transduction, cell development, and cancer occurrence.
